Brassard began his NHL career with the Columbus Blue Jackets, who selected him with the No. 6 pick in the 2006 NHL Draft after he scored 116 points (44 goals, 72 assists) in 58 games with Drummondville of the Quebec Major Junior Hockey League in 2005-06.
He played nearly six seasons with the Blue Jackets and scored 47 points (17 goals, 30 assists) in 2010-11, but was traded to the New York Rangers on April 3, 2013. He had 169 points in 309 games with the Blue Jackets.
Brassard helped the Rangers reach the 2014 Stanley Cup Final; he scored 45 points (18 goals, 27 assists) in 81 regular-season games and 12 points (six goals, six assists) in 23 playoff games. He had an NHL career-high 60 points (19 goals, 41 assists) the following season, when New York reached the Eastern Conference Final.
Brassard was traded to the Ottawa Senators on July 18, 2016. He played nearly two seasons with Ottawa before being moved again, this time to the Pittsburgh Penguins in a three-team trade that also included the Vegas Golden Knights, on Feb. 23, 2018. Brassard finished the season with Pittsburgh and scored four points (one goal, three assists) in 12 playoff games.
He started the 2018-19 season with the Penguins and had 15 points (nine goals, six assists) in 40 games but was traded to the Florida Panthers on Feb. 1, 2019. Brassard had four points (one goal, three assists) in 10 games before being traded to the Colorado Avalanche on Feb. 25. He scored four goals in 20 regular-season games and had one assist in nine playoff games.
Brassard signed a one-year contract with the Islanders on Aug. 21, 2019. He had 32 points (10 goals, 22 assists) in 66 games when the 2019-20 season was paused March 12 due to concerns surrounding the coronavirus.
Brassard signed a one-year contract with the Senators on Oct. 10, 2022, after attending training camp on a professional tryout contract. He had 23 points (13 goals, 10 assists) in 62 games was their nominee for the Bill Masterton Memorial Trophy, awarded annually by the Professional Hockey Writers' Association to the player who best exemplifies the qualities of perseverance, sportsmanship and dedication to ice hockey.
Brassard announced his retirement from the NHL on June 23, 2024, after 16 seasons.
